Survival meristem isolated from the buds of the aerial creeping shoot of garden strawberries (Fragaria×ananassa Duch.) when introduced into culture in vitro

The subject of the study is garden strawberries (Fragaria x ananassa Duch.) varieties "Mishutka", "Privlekatelnaja", "Krasnyj bereg", "Elianny", "Honey", "Elsanta", "Flora", "Albion". The aim of the study is to assess the effect of various sterilization options and types of meristem on the survival rate of strawberry explants. The research was carried out in 2018...2019 on the basis of the plant biotechnology laboratory of the Kostroma state agricultural Academy. As a result, it was found that the survival rate of explants obtained from the meristem of the apical buds of the aboveground creeping rooting shoot is 6.34% higher than from the axillary ones. The optimal duration of explant sterilization is 10 minutes. For the introduction of strawberry culture, we recommend using meristems of apical buds of creeping shoots of the donor plant, sterilized for 10 minutes in a 5% aqueous solution of the drug "NIKA-extra M", which ensured the survival rate of 73.0% of explants in vitro in the Murasige-Skuga nutrient medium with the addition of 6-BAP (0.5 mg / l), a photoperiod of 16 hours and an air temperature of 22.0 ±2.0 °C.
